Résumé

La bronchopneumopathie chronique obstructive (BPCO) est principalement associée au tabagisme. La BPCO, qui est caractérisée par le développement d’une bronchite chronique, peut être également associée à de l’emphysème. Chez les fumeurs, on observe une surexpression de cathepsine S, une protéase à cystéine, qui participerait au développement d’emphysème via son activité élastinolytique. Par ailleurs, nous avons montré que la cathepsine S dégraderait un ou plusieurs constituants protéiques des jonctions cellulaires. Cette activité protéolytique délétère conduit à une altération de l’intégrité de la barrière épithéliale pulmonaire, pouvant ainsi aggraver l’inflammation chronique et favoriser les phases d’exacerbation associées aux infections.

Summary

Smoking is accountable for most of the chronic obstructive pulmonary disease (COPD) cases. COPD, which is characterized by the development of chronic bronchitis, could be associated with emphysema. In active smokers, there is an overexpression of cathepsin S, a cysteine protease, which participates in the development of emphysema via its elastinolytic activity. Likewise, we demonstrated that cathepsin S could degrade one or more protein constituents of cell junctions. This deleterious proteolytic activity leads to an alteration of the integrity of the lung epithelial barrier, which in turn could aggravate chronic inflammation and promote the exacerbation phases associated with infections.
